Please read the DBD::Sybase manual. This issue is documented.

Because I have to open a second connection to allow you to have 2
active prepared statements I can't guarantee rollback operations
correctly (and you could very easily deadlock yourself this way too).

So AutoCommit off is not supported if you try to open more than one
$sth on a $dbh with DBD::Sybase.
