As John Yin recommended to check :

I removed J30: M0 and M2 to change the mode:

And now the memory test app works.

Memory app :

And XMD write an read are now ok:

So I continued with a pre-built design.

And now for the first time, i see the U-boot screen

So it seems jumper on J30 should be at M1 only when using DDR_RAM
without booting from Flash the design.
This might be usefull for others trying to work with the Spartan3E-500
dev board.
