You need to set that variable only at run time. That is, when you run an 
application and you want the application to link to a library that you have 
installed in non-default location. You can set this variable to point to that 
location where the library is stored before running your application.

If you are giving prefix as /usr, then your libraries will get installed in 
/usr/lib. /usr/lib is a default location and you do not need to set any 
variable.
