Thanks for the gpa-hint. I found a solution there. It really seems that  
pinentry-qt has problems (for me in combination with kde 3.1.4 not with 3.1.3). 
I changed the pinentry-program in my ~/.gnupg/gpg-agent.conf to pinentry-gtk 
and after a restart of KDE (and gpg-agent) signing mails in kmail basically now 
works again. However signed messages are marked having an invalid signature. 
I'll open another Bug report for this.
