You can do it, at least by using the gpg2.exe command line
like  
   gpg2.exe --symmetric file.txt

Decryption should work from the GUI tools like GpgEx and Kleopatra.

You can choose the algorithm also on the command line
--personal-cipher-preferences `string'
Gnupg2 supports a number of algorithms, run gpg2.exe --version
so see all supported ones.

Yes, e.g.
  gpg2.exe --symmetric --personal-cipher-preferences 'AES256' FILE.TXT
